Okay, initial thoughts here.
Firstly, you have a significant distance between the first tee and eighteenth green. Obviously you can take some liberties with it being a video game and not having to walk, and also you’re entirely free to design however and whatever you like, but for me and a sense of realism I would try to have both start and end points close to the clubhouse. It also gives you a better chance of creating some more interesting views - as it stands you’ve given a lot of real estate to a driving range and that’s pretty much all you’re going to be able to see down the right side of the first, not the most inspiring view!
Starting with a par three is quirky. I’m not a fan of it but some people and courses pull it off well - once again, you do you! In real life it can lead to pace of play concerns, but it also provides a daunting challenge when your first swing of the day has to be on the money or you’re already looking at a bogey. If that’s something you want, then go for it.
I like the first routing more, it has more variety to the hole directions. Layout two spends a lot of holes travelling diagonally from the bottom left to the top right of the plot and back again (6, 7, 8, 9, 10, 11, 13 and 18 are all broadly parallel, either into or away from the prevailing wind that you’ve indicated). It also looks so tight that often, playing down the wrong fairway would be an option.
More in depth at routing one - it looks like a lot of your holes basically play across the stream. Try using the hazard in different ways and not always having to cross the river each time; maybe have a hole run alongside it so that you can’t miss left, or a green site up against it so that it protects long?
I’m a bit confused by the “wall” you have running across the plot. I’m picturing a small stone wall, which to me doesn’t feel like it has much of a reason to be there, and also wouldn’t prevent me from cutting across much of 9, but if you can find a fun way to use it then by all means, I love new and quirky things!
If you don’t mind, I sketched a layout over the top that I think uses a lot of your ideas (half a dozen holes completely unchanged, plenty of others just shifted around a little, but I had to chuck one or two different ones in there to make my routing work) that comes back to the clubhouse after 9 and 18, and is still almost faithful to your plot (I moved the wall significantly, if you were dead keen on including one).
